INTRODUCTION
The Coenzyme Q, called also Ubiquinone (or Vitamin Q), is an organic molecule and more broadly a benzoquinone with a side chain very long isoprenic, ubiquitous in the biological systems, such as structure to the Vitamin K and to the Vitamin E.Increasing levels of Q 10 could enhance the rendement of the chain of transportation of the electrons, increasing the available energy.
There are three forms of the Coenzyme Q.
They recognize varied types of Coenzyme Q to second of the number of isoprenic unity in the side chain. In the human mitochondria the type of much more frequent CoQ is the Q 10.
The CoQ 10 amply was used by the heart treatment of dysfunctions, such as the coronary, and the cancer to the bosom.
Recent studies put in light like the antioxidizant effects of the enzyme Q 10 are able to have benefits on the body and on the brain.
EQUIPMENT AND INSTRUMENTAL PARAMETERS
The analysis of Coenzyme Q10 requires a HPLC system including isocratic pump, autosampler and UV detector.
The use of a solvent degasser prevents air bubbles and ensure a stable baseline.
Sampler: Injection volume 30 μl, analytical run time 14 min..
HPLC pump: Flow rate 1.5 ml/min is recommended
Column temperature: Ambient (~25 °C)
UV detector: 275 nm
COLLECTION AND STORAGE OF PATIENT SAMPLES
The determination of CoQ10 can be performed from whole blood, plasma, and serum.
Stored at – 20 °C the samples are stable for up to 3 months, at 4 °C for up to 3 days and at room temperature for up to 24 hours. The samples have to be stored in the dark.
SAMPLE PREPARATION PROCEDURE
500 µl Precipitation Reagent is added to vial with teflon cap
400 µl Sample is added.
50 µl Oxidation Reagent is added.
400 µl internal standard is added.
Vortex the mix for 10min.
Centrifuge at 10 000 rpm for 5 min.
Samples prepared as above can be stored at ambient temperature for about 24 hours, at +2 to +8 °C up to 36 h, frozen below -18 °C up to 3 months.
