INTRODUCTION
Urinary Formaldehyde’s dose is made after exposure to Formaldehyde and Metanol. The Formaldehyde is a pollutant so-called ubiquist, because it is everywhere, also in cities and along the very busy roads. It is one of the main pollutants present in homes. Formaldehyde is found in cosmetics, in packaging, in the glue used for books and magazines. The Formaldehyde is used in the manufacture of synthetic resins, glues, paints, solvents, deodorants and disinfectants in the industrial field. Also it is used as an intermediate in the synthesis of drugs.
The Methanol is metabolized in the liver and excreted in the urine as Formaldehyde, Formic acid and CO 2.
The important concerned effect is itsability to induce mutations and cancer. IARC (International Agency for Research on Carcer), in the month of June 2004 classified formaldehyde in Group 1 of carcinogens, namely in certain carcinogenic to humans. Thanks to this method, we can determine the urinary Formaldehyde in HPLC. It is asubstance excreted by diffusion and therefore does not depend on the concentration of creatinine.
EQUIPMENT AND INSTRUMENTAL PARAMETERS
The analysis of 2,5 -hexanedıone requires a HPLC system including isocratic pump, autosampler and UV detector.
The use of a solvent degasser prevents air bubbles and ensure a stable baseline.
Sampler: Injection volume 50 μl, analytical run time 4 min..
HPLC pump: Flow rate 1.2 ml/min is recommended
Column temperature: Ambient (~25 °C)
UV detector: 352 nm
COLLECTION AND STORAGE OF PATIENT SAMPLES
After work shift, collect 10 ml of urine in a suitable test tube.
The sample is stable up to 4 weeks at - 20 °C.
SAMPLE PREPARATION PROCEDURE
50 µl urine sample is added to vial with teflon cap.
100 µl hydrolysis reagent is added.
100 µl Buffer Solution is added.
100 µl Derivatization Solutionis added.
Vortex the mix for 10s.
Incubation is carried out at 70°C for 15 min.
500 µl of H2O HPLC grade is added.
Samples prepared as above can be stored at ambient temperature for about 24 hours, at +2 to +8 °C up to 36 h, frozen below -18 °C up to 3 months.
